Pharmacology and Toxicology: Gefarnate protects the gastric mucosa, promotes ulcer healing, increases gastric mucosal prostaglandins, prevents a decrease in mucosal electrical potential, promotes soluble mucus secretion, increases visible mucus layer thickness, strengthens the gastric mucosal barrier, dilates gastric mucosal microcirculation, and improves blood flow distribution.
Pharmacokinetics: Absorption, Distribution, and Excretion:
A single oral dose of 50 mg/kg of gefarnate showed an absorption rate of 60-70%. Peak blood concentrations reached 6 hours after oral administration. After 24 hours, 12.4% was excreted in urine, 19.5% in expired air, and 30-40% in feces.
