[Indications]
Threatened abortion and habitual abortion, premenstrual tension syndrome, anovulatory functional uterine bleeding and anovulatory amenorrhea, combined with estrogen to treat menopausal syndrome.
[Usage and Dosage]
1. Combined with estrogen (such as conjugated estrogen, Premarin): Premarin tablets: oral, 1.25 mg, once a day, for a total of 22 days; take this product from the 13th day of taking Premarin tablets, oral, 200 mg (4 tablets), twice a day, for a total of 10 days.
2. For threatened abortion and habitual abortion, premenstrual tension syndrome, anovulatory functional uterine bleeding and anovulatory amenorrhea, the conventional dose is 200mg-300mg (4-6 tablets) per day, taken once or twice. Each dose should not exceed 200mg (4 tablets), and the medication time is best to be far away from meal time.
[Adverse Reactions]
Breakthrough bleeding, vaginal punctate bleeding, weight gain or loss, changes in the cervical squamous-columnar junction, changes in the characteristics of cervical secretions, breast swelling, nausea, dizziness. Headache, fatigue, fever, insomnia, allergies with or without itching, melasma, chloasma, obstructive jaundice, abnormal liver function.
[Contraindications]
1. Allergic to progesterone or other ingredients in this product; 2. Vaginal bleeding of unknown cause; 3. Thrombophlebitis, vascular embolism, cerebral stroke or previous medical history; 4. Breast tumor or genital tumor.
【Precautions】
1. Patients with kidney disease, heart disease, edema, and hypertension should use with caution. 2. Once clinical manifestations of thrombotic diseases (such as thrombophlebitis, cerebrovascular disease, pulmonary embolism, retinal thrombosis) appear, the drug should be discontinued immediately. 3. Sudden partial vision loss or sudden blindness, diplopia or migraine should be discontinued immediately.

【Drug interactions】
Drugs such as phenobarbital, phenytoin sodium, and rifampicin can weaken the efficacy of this product because they have an induction effect on cytochrome P450. Ketoconazole is an inhibitor of cytochrome P450 enzymes. Therefore, ketoconazole or other cytochrome enzyme inhibitors may increase the blood concentration of progesterone.
[Pharmacological action]
Progesterone is a steroid hormone secreted by the ovaries, placenta and adrenal glands. In the presence of sufficient estrogen, progesterone can change the endometrium from the proliferative phase to the secretory phase, providing favorable conditions for the implantation of the fertilized egg. After the fertilized egg implants, the placenta is formed, which can nourish the excitability of the pregnant uterus and maintain the pregnancy state; it can promote breast development and prepare for breastfeeding. This product can inhibit the release of luteinizing hormone from the anterior pituitary through negative feedback to the hypothalamus, so that the follicles cannot mature and inhibit the ovulation process of the ovaries.
